瘦素(leptin)是由ob基因编码、在脂肪组织中生成的一种脂肪代谢调控产物，在代谢和调控体重等方面发挥重要作用。它通过下丘脑中的瘦素受体发出信号，降低食欲，增加能量消耗。在外周组织中，瘦素能拮抗胰岛素信号传导，增加脂肪酸氧化，减少胰岛β细胞的胰岛素分泌，并促进生育。它在脂肪细胞中的表达可以被其他脂肪因子如il-6（刺激）和tnf-α（抑制）以旁分泌方式调节。瘦素缺乏的小鼠表现出严重的胰岛素抵抗、肥胖和生育能力下降，所有这些都可以通过外源性瘦素给药而逆转。还有研究表明，瘦素与心血管疾病有关，因为它介导了许多促炎和促动脉粥样硬化作用，如内皮素-1（et-1）表达增加、活性氧（ros）积累、单核细胞趋化蛋白-1（mcp-1）分泌以及交感血管张力和血压增加。
